To kill two Pinkertons during the ambush you simply need to tag several before you start firing, it's quite easy to get 4 or 5 of them however. Then when on foot you don't need to worry about time, accuracy or headshots at all. Then when you get on the Gatling Gun, use your Dead Eye to fire single shots at the head. Once you've got 5 headshots, plus maybe an extra two to be certain, go for body shots to be sure of the accuracy stat. If you approach the mission like that, you should have no problem.
