I like to take out the two agents downstairs closest to you, going after the one upstairs and outside may take too much time, and they won't be able to see you leave anyway. A few other tips with the time - once you are riding on the route, you can switch the cinematic to put your horse on auto pilot to make it to Shady Belle. Also the letter is always in the same place, you don't need to bother inspecting the other clues, so go straight for it. For the record, cutscenes were skipped in my play through, and edited back in later, although there is debate about whether the timer pauses or not for all missions. This one however, you should be able to beat the time comfortably even the cutscenes do count to the time and you don't skip them!
